Sample extraction:
1. For seed extracts: Imbibe seeds overnight in water.
Remove the seed coat and put each seed to be tested in a microcentrifuge tube. Add 500ul-sample extraction buffer. Crush with a pestle for 30 seconds. Centrifuge for 60 sec in a microfuge. Place the Xpresstrip in the extract.
2. For leaf extracts: Punch out 4 leaf discs with a microcentrifuge tube by placing a leaf
between the lid and the tube opening and closing the lid onto the leaf. Add 500ul-sample
extraction buffer. Crush with a pestle for 30 seconds. Place
